Feasibility consultation

A feasibility consultation is always free!  If you’re new to cryo-EM, we’ll gladly walk you through the basics and determine if single particle analysis is the right method for your sample.

Cryogrid sample preparation

Send us your purified sample and we’ll prep your cryogrids at the same facility where we conduct screening and data collection.  We’ll store them as long as you need, and can also facilitate shipping of grids from one site to another for screening or data collection.

Screening and Data Collection

Once your cryogrids are made, we’ll schedule time on one of our partner facility cryo-TEMs for an initial screening and potential data collection.  

preliminary single-particle analysis

For those still working on sample optimization, this service is an analysis of a small dataset (<500 movies) to determine if the sample is suitable for high-resolution single-particle analysis.  We’ll run 2D classification, return a low to moderate resolution structure and provide an estimate of how much data you’ll need to achieve your structure goals.

Full Single-Particle Analysis Workflow

Once you’ve collected a full cryo-EM dataset, we’ll get to work on data analysis and 3D reconstruction.  Some protein samples can be resolved to high resolution in just a few days, while others can be more difficult and require longer processing times.  For extremely difficult or mobile samples, we offer Advanced Single-Particle Analysis, which can often take several weeks.

Advanced single-particle analysis

For those working with very difficult proteins.  Some samples are very heterogeneous or have mobile domains that can be difficult to resolve with a standard data analysis workflow.  Subunit masking, non-uniform refinements, and other tools can help increase resolution of these samples.  This level of data analysis can take several weeks.

Model Building

After generation of a high-quality 3D reconstruction, you’ll have the option to build an atomic model into your newly acquired density map.  In addition to a full statistical report on the quality of the model, we’ll also assist you in creating any figures you may need for publication, advertising, or just for showing off to your colleagues.
